Recent progress in the chemical synthesis of proteins

Curr Opin Struct Biol. 2014 Jun:26:16-23. doi: 10.1016/j.sbi.2014.03.002. Epub 2014 Mar 27.

Abstract

A methodology for chemical protein synthesis by ligation chemistry has become increasingly practical and applied to the synthesis of various proteins and glycoproteins for their structural and functional analyses. This is largely due to the advances in the synthesis of peptide thioester as well as to the progress in the extension of the ligation chemistry to a non-cysteine site, which are briefly summarized in this review.
