A mutational study of a Diels-Alderase catalytic antibody

Abstract

Site-directed mutagenesis was used to probe the mechanism of antibody 39-A11, which catalyzes the Diels-Alder reaction of substrate 1 and 2 to give cycloadduct 3. Mutations that lead to improved packing interactions with the diene afford an order of magnitude increase in catalytic activity.
